Efflux Pumps and Multidrug-Resistance in Pyricularia oryzae Triticum Lineage

چکیده
Widespread resistance to QoIs, DMI and SDHIs fungicides has been reported for Brazilian populations of the wheat blast pathogen Pyricularia oryzae Triticum lineage (PoTl). A pre-existing mechanism not associated with target site mutations indicated DMIs SDHIs, strong indication that PoTl multidrugresistance (MDR). Therefore, main objective this study was test hypothesis SDHI detected in due efflux pump mediated MDR mechanism(s) by characterizing sensitivity antifungal substrates. Four substrates were tested: tolnaftate (TOL), cycloheximide (CHX), rhodamine 6G (RH6G) triphenyltin chloride (TPCL). TPCL RH6G considered most relevant indicators enhanced activity. Among 16 isolates tested, 9 insensitive TPCL, 1 TOL, CHX. The grouped into four distinct multidrug phenotypes (MDRPs) based on combinations Insensitivity or TOL correlated well insensitivity, but resistance. identification multiple MDRP our warrants further research aimed at revealing exact mechanisms pathogen, including pumps overexpression via transcriptomic analyses differentially expressed genes; discovery changes promoter regions transcription factors transporters

Porins, efflux pumps and multidrug resistance in Acinetobacter baumannii.
Acinetobacter baumannii is an opportunistic pathogen, causing infections mainly in patients in intensive care units where the extensive use of antimicrobial agents can select for the emergence of multiresistant strains. In fact, since strains resistant to all antimicrobial agents have been reported, A. baumannii is considered the paradigm of multiresistant bacteria. Both acquired and intrinsic ...

OBJECTIVES salmonella enterica strains exhibiting decreased susceptibility to tigecycline have been reported. In this study, we sought to elucidate the roles of Salmonella multidrug efflux pumps and AcrAB regulators in tigecycline resistance. METHODS we examined the involvement of multidrug efflux pumps and AcrAB regulators in resistance to tigecycline and other glycylcyclines by determining ...
متن کاملClinically relevant chromosomally encoded multidrug resistance efflux pumps in bacteria.
Efflux pump genes and proteins are present in both antibiotic-susceptible and antibiotic-resistant bacteria. Pumps may be specific for one substrate or may transport a range of structurally dissimilar compounds (including antibiotics of multiple classes); such pumps can be associated with multiple drug (antibiotic) resistance (MDR). However, the clinical relevance of efflux-mediated resistance ...
متن کاملMultidrug Efflux Pumps of E. coli 215 AcrAB and Related Multidrug Efflux Pumps of Escherichia coli
The AcrAB system of Escherichia coli is a multidrug efflux system composed of an RND-type transporter AcrB and a periplasmic accessory protein AcrA, and pumps out a wide variety of lipophilic and amphiphilic inhibitors directly into the medium, presumably through the TolC outer membrane channel. AcrA, a highly elongated protein, is thought to bring the outer and inner membranes closer. It forms...
متن کاملBacterial Multidrug Efflux Pumps: Much More Than Antibiotic Resistance Determinants
Bacterial multidrug efflux pumps are antibiotic resistance determinants present in all microorganisms. With few exceptions, they are chromosomally encoded and present a conserved organization both at the genetic and at the protein levels. In addition, most, if not all, strains of a given bacterial species present the same chromosomally-encoded efflux pumps. Altogether this indicates that multid...
